Concomitant administration of VC and EtOH also produced an excess of hepatocellular carcinoma and lymphosarcoma. EtOH with or without VC had a strong tumorigenic effect on the endocrine system. Most of the angiosarcomas were highly anaplastic, primary tumors in the livers that metastasized to the lungs. Only 1 angiosarcoma was seen in all the control rats; that occurred in s.c. tissue. The neoplastic cells were transformed from type II alveolar epithelium via its hyperplastic form. Nonneoplastic effects of the chem. were also studied in the 27 mice. Major light microscopic alterations obsd. were proliferation and hypertrophy of the terminal bronchiolar cells, consisting of ciliated and Clara cells, hypersecretion of the epithelial mucin in the goblet cells of both the bronchial and the proximal bronchiolar epithelium, hyperplasia of alveolar epithelium, mobilisation of alveolar macrophages, and occasional presence of peri bronchial or bronchiolar chronic inflammation. Submicrosccpic changes of pulmonary alveoli were represented by focal thickening of the basement membrane, multiple foci of hyperplastic type II cell (the precondition of the alveologenic tumor), active discharge of osmiophilic lamellar bodies from the type II cell and phagocytosis of the bodies by macrophages, appearance of cholesterol crystalloids in the macrophages, degeneration of alveolar septal cells, and occasional appearance of a large nucleus with swelling of the capillary endothelium. The neoplastic effect of vinyl chloride of smaller doses C100, 10, 1 and 0 (control) ppm] and shorter exposure (4 wk) was studied in lungs of 120 mice. 2000 ENG AJIMD 0271-3586 CBAC COPYRIGHT: CHEM ABS Whether vinyl chloride monomer (VCM) (75-01-4) or 1 of its metabolites, chloracetaldehyde (107-20-0), has a short-term effect on cell metab. was studied. ATP (56-65-5), ATP (58-64-0), And AMP (61-19-8) levels were measured in regenerating rat liver after 4 and 16 h. No changes in the levels of these nucleotides was evident after i.v. injection of VCM immediately after partial hepatectomy. A 2nd technique, that of measuring transmembrane potentials with microelectrodes, was applied both to cells in rat liver slices and pulmonary alveolar macrophages. The addn. of VCM and chloracetaldehyde with the electrodes in position in rat liver slices had no significant effect on transmembrane potentials. The same was true for a series of measurements on macrophages before and after addn. of the test chem. It therefore appears that VCM does not alter general cell metab. but could act on a specific intracellular target.

Exposure of male mice to different chems. altered the transformation characteristics of their splenic lymphocytes. Addn. of chems. directly to lymphocyte cultures also caused different effects indicating that some of the effects can be caused directly on the cells. Thus, lymphccytv. cultures from spleens can be utilised for toxicity evaluation of chems. on immune functions of the organism.
