METHOD:________________________________________________________ Method: ASTM E1367-90, Standard guide for conducting 10 day static sediment toxicity tests with marine and estuarine sediments. Type: Acute static GLP: No Year completed: 2000 Species: Corophium volutator Supplier: Field collected from an unpolluted site near Trondheim, Norway. Analytical monitoring: pH and dissolved oxygen. Exposure period: 10 days Test organism age: Adults (4-8 mm size) Statistical method: LC50 values calculated by PROBIT Analysis, National Swedish Environmental Protection Board (Version 2.3,1990). Calculation of NOEC by single-factor ANOVA followed by Bonferronis procedure. Test conditions: Dilution water: Natural seawater (34 ppt salinity) from an unpolluted site in Byfjorden outside of Stavanger, Norway. Sediment collected at an unpolluted site. The sediment is a muddy sand and had an organic fraction of 0.7% and a silt fraction of 0.8%. Dilution water chemistry: Not given. Lighting: Not given. Stock and test solution preparation: Each test concentration was prepared by adding the test substance to 600 g damp sediment and 150 mL of seawater, then shaken for 3 hours. The sediment was split into three subsamples, with an overlay of 400 mL seawater added to each, and aerated overnight before study initiation. 002407 Exposure vessels: 600 mL glass beakers containing 200 g of test sediment and ~400 mL of seawater with continuous aeration. Number of replicates: 3 Number of organisms per replicate: 10 Number of concentrations: four plus a blank control Water chemistry during the study: pH range (0-10 days): 8.1 - 8 . 1 (control exposure) 8.0 - 8.1 (3060 mg/kg exposure) Temperature range: 15 1 C Dissolved oxygen range (0-10 days): 90 - 97% (control exposure) 93 - 100% (3060 mg/kg exposure) RESULTS___________________________________________________ Nominal concentrations: Blank control, 222,457,1060, and 3060 mg/kg (dry weight). Reference substance (fluoranthene) at 20.9 mg/kg dry weight. Element values: 10-day LC50 = >3060 mg/kg (sediment dry weight) 10-day NOEC = 3060 mg/kg (sediment dry weight) Element values based on nominal concentrations Remarks: Testing was conducted on the mixture as described in the Test Substance Remarks field. The values reported apply to that mixture and not the fluorochemical proportion alone. CONCLUSIONS _________________________________________________ The test sample 10-day LC50 was determined to be >3060 mg/kg (sediment dry weight). The 10-day No Observed Effect Concentration (NOEC) was 3060 mg/kg (sediment dry weight). The 3060 concentration was the highest concentration tested and caused ~17% mortality. Validity criteria fulfilled and reference substance obtained mortality of 16.7%. Submitter: 3M Company, Environmental Laboratory, P.O. Box 33331, St. Paul, Minnesota, 55133 DATA QUALITY___________________________________________________ Reliability: Klimisch ranking = 2. This study meets the criteria for quality testing. However, sample purity was not properly characterized and the study lacks analytical confirmation of the amount of fluorochemical proportion in the solution. 002408 REFERENCES Test was conducted by RF Rogaland Research, of Stavanger, Norway at the request of 3M Norge, Lab reference number 99312-4, 2000. TEST DETAILS: METHOD: NS-EN ISO 10253,1998: Water quality - Marine algal growth inhibition test with Skeletonema costatum and Phaeodactylum tricomutum. TEST ORGANISM: Skeletonema costatum, strain NIVA BAC 1. A stock culture o f the test p organism is kept at RF - Rogaland Research. PREPARATIONS: Natural seawater (salinity: 34 %o) from an unpolluted site in Byfjord was used as basis for the growth medium. The water was filtered through a GF/C filter, and briefly heated to about 75C prior to use. The seawater was enriched with nutrients according to ISO 10253, and recommendations from the Paris .. Vn Com m ission. i Test solutions were prepared from a stock solution. A stock solution o f 1020 mg/1 was prepared by stirring 1.020 grams o f the product in 1 litre o f growth medium. Other concentrations were prepared from this stock solution. INCUBATION: Test cultures o f 50 ml were incubated in 100 ml flat-bottomed flasks on an orbital shaker, 3 replicates for each concentration. Test cultures were inocu lated to about 2000 cells/m l from an exponentially growing inoculum culture^ The test was run under continuous light (> 50 pE/m2-sek) from fluorescent^-- tubes o f universal natural white type, at a temperature o f 20 2C. The test was run for about 72 hours, and subsamples o f 5 ml were taken from the test flasks each day for growth measurements. Algal growth was measured by fluorescence. pH measurements were performed as indicated on data sheet. 00241 Lab.ref.no.: 99312-4 Page 6 o f 31 DATA ANALYSIS: CALCULATIONS: Growth rates were calculated by log linear regression on biomass (fluorescence) versus time, as described in NS-EN ISO 10253. ErCso: The inhibitory effect on the growth o f Skeletonema costatum is expressed as the concentration o f a product giving 50% reduction of growth rate (ErCso). This effect concentration was calculated by interpolation between 100mg/l and 320m g/l. NOEC: Calculation o f NOEC (No Observed Effect Concentration) is done by single factor ANOVA follow ed by Bonferronis procedure for comparing each concentration mean with the control mean. TEST RESULTS: Table 1. EC and NOEC values obtained with Skeletonema costatum. Test substance "F C -603 EF Light Water TM A T C TM 3%" ErCio - EfCso -170 EAo 100<EC9o<320 COMMENTS: NOEC: 100 Values indicate concentrations giving 10%, 50% and 90% inhibition o fgrowth rate. Units in mg/L Values are calculated by interpolation. NOEC are calculated by using the computerprogramme GraphPad InStat 3.0. The results are based on growth rates for the time span 0 - 72 h, the period during which the growth is considered to be exponential in the control cultures (see growth curves). VALIDATION: The validity criteria given in the test protocol are fulfilled, in that the control cell density increased by a factor o f more than 16 in the test period (corresponding to a growth rate o f more than 0,9 d'1), the control pH did not vary by more than 1,0 units during the test and the 72 hours test results with 3,5-dichlorophenol was in the range normally found.
