Local Lymph Node Assay Screen with H22051
> f l % The Local Lymph Node Assay (LLNA) screen with H22051BfUwas conducted
in female CBA/JHsd mice. Experimental details of the stuudy are located in laboratory
notebook E87092.
The test substance was prepared in 2:1 dimethylsulfoxide (DMSO)/acetone and administered topically to the dorsum of both ears of mice (6 per group) at dosages of 0% (DMSO/aceione vehicle), 1%. 10%, 25%, 50%, and 100%. An additional group of 6 female mice was dosed with 25% hexylcinnamaldehyde (HCA) as a positive control.
Clinical observations made on day 5 indicated alopecia in the neck area in 5 of 6 mice at
the 100% concentration.
No statistically significant differences in the mean body weights or mean body weight gains were observed at any concentration. All cell proliferation measurements at all concentrations of the test substance produced no statistically significant differences. In addition, all stimulation indexes were less than 3.0 and no dose response was present. Under the conditions of this study, H22051 wa^ not a sensitizer. It was also concluded that a 25% concentration of HCA produced a positive sensitization response in mice,
