Thank you very much indeed for your cooperation in this research pro ject. I have talked with Mr. Hugh Reid, Administrator of our laboratory, about payment of the inhalation experiment cost, and he has told me that there is no problem for this matter. I understand that this new inhalation will be initiated by the end of this month (October) and the animals will be shipped to New York by airplane by the end of November (27th or 28th), immediately after the one month exposure. I do not have any problems with this schedule. If some of the animals are found dead during the process of the exposure, please fix those animals by a perfusion* of 10% buffered neutral forma lin via Inferior Vena Cava or Aorta, without opening the chest, before major organs are taken out from the bodies. After perfusion, major organs such as lung, liver, brain, stomach, kidneys, testis, spleen and any abnormal organs should be refixed in the fixative. Later, those organs and the bodies should be sent to New York for further pro cess of histological studies. If some animals are found fatally sick, the animals should, be sacri ficed by a deep anesthesia of ether**, then a perfusion of paraform aldehyde without sugar*** should be performed as pre-fixations for light and electron microscopy. For electron microscopy, lung (right lower lobe; 15 blocks), liver (5 blocks), brain (occipital area; 5 blocks) and abnormal tissue should be kept in the paraform aldehyde fixative for further steps in New York. For light microscopy, after the pre-fixation by paraform aldehyde, the major organs stated before should be post-fixed in 10% neutral buffered fixative to be shipped to New York with the bodies. Therefore, at this time, please set a lower position (about 1 meter in height) of the fixative bottle. **To avoid structural damages caused by the perfusion under anesthesia, a complete death of the animals may be necessary. ***The previous E.M. fixative was "hypertonic", and there was some diffi culties in staining of light microscopic sections, possibly due to existence of sugar in the fixative.
