NS-EN ISO 10253, 1998: Water quality- Marine algalgrowth inhibitiontest with Skeletonet?icaostalum and Phaeodactylum tricomutum.
TEST ORGANISM: Skeletoneilicaovtatum,strainNIVA BAC 1. A stockcultureof the test organism iskept at RF - Rogaland Research.
PREPAMTIONS:
Natural seawater (salinity':-'%)o4) from an unpollutedsiteinByfjord was used as basisforthe growth medium. The water was filteretdhrough a GF/C filter, and brieflyheated to about 75'C priorto use. The seawaterwas enrichedwith nutrientsaccording to ISO 10253, and recommendations from theParis Conunission.
Test solutionswere prepared from a stock solution.A stock solutionof 511.2 mg/l was prepared by stirrin0g.5112 grams of the product in I litreof growth medium. Other concentrationswere prepared from thisstock solution.
INCUBATION:
Test culturesof 50 ml were incubatedin 100 ml flat-bottomedflaskson an orbitalshaker,3 replicatesforeach concentration.Test cultureswere inoculatedto about 2000 cells/mifrom an exponentially1_?owing inoculum culture. The testwas run under continuouslight(> 50 gE/m Sek)from fluorescent tubes of universalnaturalwhite type,at a temperatureof 20 2*C.
The testwas rL7-fnor about 72 hours, and subsamples of 5 ml were taken from thetestflaskseach day forgrowth measurements. Algal growth was measured by fluorescence.pH measurements were performed as indicatedon data sheet.

ASTM E1367-90, Standard Guide For Conducting 10 day StaticSediment ToxicityTestswith Marine and EstuarineSediments.
TEST oRGANism: Corophium volutator,ftom an unpollutedsitenear Trondheim.
CULTIVATION:
The testorganisms were suppliedas adults(4-8mm in size)and were held in aeratedseawaterat 15'C. A small amount of naturalseaweed was used for feeding.
PREPARANON:
Natural seawater(salinity3:4 %o) from an unpollutedsitein Byfjorden outside Stavanger was used fordilutionwater. Sediment collectedatan unpolluted site was used forpreparing testsediments. The sediment isa muddy sand and was analysed fororganic contentand proportionsiltforeach test.
Each testconcentrationwas prepared by adding the product in 600 grammes of damp sediment in a plasticbucket. 200 ml of seawaterwas added and the bucket was shaken for3 hours. The sediment was divided intothreebeakers, 400 ml of seawater was added to each,and the beakers were aeratedover night. 10 animals were then randomly selectedfor each beaker.
