CHRONIC TOXICITY TO FRESHWATER INVERTABRATES (Daphnia magna)
TEST SUBSTANCE
Identity: Perfluorooctanoic acid, ammonium salt; may also be referred to as PFOA ammonium salt, Ammonium perfluorooctanoate, FC116, FC-126, FC-169, orFC-143. (Octanoic acid, pentadecafluoro-, ammonium salt, CAS # 3825-26-1 )
Remarks: The 3M production lot number is 264. The test sample is FC143. It's purity was not sufficiently characterized, though current information indicates it is a mixture of 96.5 - 100% test substance and 0 3.5% C6, C7, and C9 perfluoro analogue compounds. The 3M production lot number is 264.
METHOD:
Method: USEPA-1982, OECD 1981. Type: Semi-Static Life-Cycle Toxicity GLP: No Year completed: 1984 Species: Daphnia magna Supplier: In-house cultures, 3M Environmental Lab, St Paul, MN Analytical monitoring: Temperature, pH and dissolved oxygen Exposure period: 21 days Statistical methods: Probit analysis (for acute toxicity values) and Moving Average Angle Method for chronic study. Statistics were recalculated by AScI Corp., Duluth, MN in 1998. NOECs and LOECs were generated using Toxstat. Reproduction was normal and
homogenous therefore Dunnett's Test was used. Fisher's Test was used for survival. IC50S (reproduction) were generated using ICp program. Survival EC50S were generated using Trimmed spearman-Karber. Test organism age: < 24-hour old neonates for test initiation. Pretreatment: None
Test conditions:
Dilution water: Aerated carbon-filtered well water
Dilution water chemistry (sampled one month prior to study
initiation)
hardness: 240 mg/L as C aC 03
alkalinity: 230 mg/L as C aC 03
pH: 7.8
Conductivity: 340 pmhos/cm
COD:
< 0 .4 mg/L
Stock and test solution preparation: Information not available
003824
Renewal rate: Every two days Exposure vessels: 250-mL glass beakers containing approximately 200 mL test solution. The depth was approximately 5 cm. Beakers were covered with watch glasses Number of replicates: 4 Number of test organisms per replicate: 5 Loading: 1 daphnid per 40 mL Number of concentrations: 6 plus a blank control
Feeding: A suspension of fish food (trout chow) and yeast containing 5 mg dry solids per one mL of mixture was fed on a daily basis Lighting: Cool-white fluorescent lights. Intensity ranged 50-100 ftc. Photoperiod of 16-hours light, 8-hours dark with a 20-minute transition period^
Water chemistry of new and old solutions during the study: PH and dissolved oxygen: Information not available Test temperature range ( 0 - 2 1 days): 2 2 + 2C
Element basis: Survival and reproduction.
RESULTS
Nominal concentrations: Blank control, 5, 8, 13, 22, 36, and 60 mg/L Element value: 14-day NOEC (survival) = 60 mg/L
14-day NOEC (reproduction) = 8 mg/L 21 -day NOEC (survival) = 22 mg/L 21-day NOEC (reproduction) = 22 mg/L 14-day IC50 (reproduction) = 40 (28-48) mg/L 21-day IC50 (reproduction) = 43 (35 - 46) mg/L
All element values based on mean measured concentrations
Biological observations Survival: All surviving first generation daphnids appeared normal at test termination. Survival in the 36 and 60 mg/L treatments was statistically significantly different from the negative control group. Reproduction: Daphnids in the control started producing neonates on Day 7. Reproduction was statistically significantly different from the control (Dunnett's) at the 13, 22, 36, and 60 mg/L concentrations after 14-days, and in the 36 and 60 mg/L test solutions at 21-days.

*Final results are calculated from mean values of cumulative no. young per adult, or cumulative mortality of adults.
Definitions ;
Daphnia Chronic Toxicity Test - A 14-21 day experimental study of the survival, growth and reproduction of asexually reproducing Daphnia beginning with- 12+12 hr old organism.
Significant - "Statistically significantly different" refers to p<0.05 with respect to the control.
Approximate NOEC - The highest test concentration that will produce no significant observed effect on reproduction of young or adult survival.
* Chronic EC5Q(s) - The values of toxicant concentrations which impair reproduction of young or induce adult mortality by 50%.
MATC - The highest concentration of toxicant that has no adverse effect on survival, growth, and reproduction of a species.
GM-MATC is the calculated geometric mean between the highest test concentration having no significant effect and the lowest test concentration having a significant effect.
Application Factor (AF) - The quotient of the chronic MATC divided by the acute 48-hr EC5q (MATC/Acute EC^g). "AF" is a number used to estimate concentration of toxicant that will not cause significant effect to a population during chronic exposure.
Environmental Safety Factor - The ratio of the chronic MATC with the expected environmental concentration (MATC/EECf. Safety factors >20 at the predicted exposure level are desirable to have sufficient confidence in the environmental safety assessment.

Dear Sue:
FC-143 Daphnia magna reproduction data for concentrations above the survival NOEC were not included during hypothesis testing for the reproduction NOEC and LOEC. EPA guidance stipulates that concentrations which significantly effect survival should not be included in hypothesis testing for reproduction. The exclusion of such concentrations usually results in a more powerful and appropriate statistical analysis.
