To determine the irritating, fatiguing and sensitizing properties of the test article. The method em ployed in carrying out this test w as sim ilar to that described in "Appraisal of the Safety of Chem icals in Foods, Drugs and Cosmetics" by J.H. Draize and published by the Association of Food and Drug Officials of the United States. The test involved the application o f the test article to the upper arms o f a group o f 113 volunteer subjects. O ne hundred and six subjects completed the study (see Subject List). These subjects w ere all determined to be in good general health and free o f any visible skin disease or anom aly in the area to be patched. Each subject w as required to read, understand and sign an inform ed consent statem ent. The patch w as then applied to th e contact site and rem ained in place for 24 hours. A t the end o f this period, the patch w as rem oved and the site was exam ined for any dermal response. Scoring o f irritation w as in accordance w ith d ie scale in the Appendix. The subjects w ere then rested for a 24 hour period after which the skin site w as again exam ined. A patch w as then applied to die sam e site as previously used. The second application w as idenfical to the first and rem an ed in place 24 hours. This procedure w as repeated on M ondays, W ednesdays and Fridays until a series of nine applications had been m ade. Patch applications m ade on Friday were rem oved by d ie subject on Saturday. The subjects exam ined the site for any dermal response and reported their observations prior to the next application. The sam e site continued to be used throughout the study. Challenge Phase After the 9th application, a rest period of approxim ately 2 w eeks elapsed after w hich a challenge application w as applied in the sam e m anner and to the sam e site described above. A duplicate challenge application w as J also applied in d ie sam e manner to a virgin site (previously untreated) on the other arm. The challenge application w as rem oved after 24 hours and the sites w ere exam ined for signs of irritation or sensitisation. Follow up exam inations w ere conducted at 48 hours after the challenge application. The subjects involved in this study ranged in age from 16 to 67 years of age. Results. See accom panying table, summary and appendix. It differs from primary irritation in that it m ay also involve tissues remote from the site o f application. These reactions represent an allergy to a chemical w hich is not obtained on first contact, but requires a num ber o f prior exposures for developm ent. Repeated patch applications of substances to both man and anim als reveal that certain substances w hich, at a given concentration, do not produce primary irritation m ay e^ dt severe skin reactions after a number o f exposures. N evertheless, these reactions may not be considered incidents o f sensitization since after 10-14 days of rest the skin reo vers its original resistance to injury by the substance. For fire lad; o f a better term such reactions have been called "skin fatigue". This latter term is a m isnom er in that it does not im ply a condition in w hich the skin has lost significant norm al physiological function.
