INTRODUCTION An experiment has boon in progress at The Saranac laboratory for somo e 14 months in an attempt to ascertain the influence of inhaled asbestos duct oe upon the incidence of pulmonary tumors in mice The purpose of this study is to obtain oxporituental data for correlation with experience portaining to pulmonary cancer in industrial workers exposed occupationally to the inhalation inhalation inhalation of asbestos dust . Two strains of mice one susceptiblo and the other resistant to the development of pulmonary tusors have been exposed to the acbestos dust comparable numbers of control animals of each strain have been kopt in normal air but under conditions of housing and dict identical with those of the experimental amirals The experiment has been designed to reveal a possible carlier onzot of tumors in the exposed animals than in the control group as well as to determine the total incidence of tumors occurring ovor + the entire poriod of the experiment 2 It may be stated lore that the evidence eae ne ere oo 2 obtained reveals were no tondency awe we , towards an carlier onset of tumors among exposed animals in comparison with TT re wena! - control animals control Nevertheless there does ~e" ee appear to be a trend toward a oe greator incidence of tumors in the exposed arimals than in the control animals TT NN ee wets -- ae ae It is important to note however that statistical analysis of the data obtained up to the pro.ont shows that the difforeness bobeen the oxpori- montal and tho control animals are no greater than might be expected to cccur by chance alone The experiment is not completed and it likely that if the ratios of tumor incidence at the next period when animals i . of 4 . b . ae e = ) tyros tee re LES en bue, Tyce 1st 23 5-7-53 - 11 stst 1st LM! jee anReport Report Report 5-7-53 5-7-53 5-7-53 . are killed killed killed are skilar to thone for tho is now notod may finally be shot to to month intorvel tho trond which statistically statistically significant because of the larger total number of animals observed . The observations made at cach of the periods when animals were killedkilled periods are presented in table 1. It will be noted that the total number of animals examined in cach of the different groups varies slightly This ver^>ationver^>ation in the result of a few spontaneous deaths in each group The observations nade in such cases harte beon incorporated in the data for the next period In many instances animals dying spontaneously have toon eaten by their cagemates It has bosh necessary to discard those animals completely from tho record of experimental observations It will be noted blat within Strain G there is a higior percentage of tumors among animals exposed to asbostes dust then among the controls The samo situation is true within the Strain 057 black Obviously the same condition is true also of the overall figures combining both strains The significance of all those porcentage incidence figures has been tested statistically by means of the recopied formula S.E. oo Standard orror of difference in incidence between two groups Total number of animals in the grov^ j _ 0 ay ~ wtiwbe 425001 1st1st1 rim Report Report 5-7-52 P1 = Per cent of animals with tumor in a specific group of animals * * 8 " Qs without tumor in the same specific group Thus Q1 = 100 ) Ya F2 02 Corresponding symbols for another group of animals It will be recalled that if an observed difference in porcentage incidence between two groups is greater than twice the standard error of that difference the difference may beconsidered to be significant and due to factors other than chance If the observed difference is less than twice the standard error it is very likely due simply to chanco In table2 the observed difference in the incidence of pulmonary tumor and the critical value of twice the standard error of difference are compared for the various groups I will be noted by comparing the values that none of the observations nodo to dato tre statistically significant An example of the method of calculation follows Calculation of standard error of difference between over incidence of tumors in the control animals and in the experimental mice from data in tablo 1 : Incidonco of turiors poriental animals 9.5 Control anuals 5.5 Number of animals observed Experimental animals 379 Control animals 1.31 S.E. \r9.5 . : x 909 .5 4 5.5. a 5 94.5 > 1.79 381 2.77 4.80 2.87 4.80 4.+80 2.87 * 17.07 17.07 : CO . . ; ~ Mining sbestos Assoc 1st Lrin Report 5-7-52 Thus a difference up to 5.5 per cent is likely to occur by chance The observed difference of 1 per cent 9.5-5.5 9.5-5.5 9.5-5.5 = 4.0 may therefore have occurred simply by chance distribution of enimals enimals and is not statistically significant V. CONCENT It will be apparent from a review of table 1 that the experiment is now in its most important phase The incidence of pulmonary tumors has begun to rice for animals killedat control animals and for the animals the month period both exposed to asbestos dust for tho The incrcos incidence is moro pronounced for the group exposed to dust but the rise has occurred at about the same time in both groups The differences betwoon the two groups however as has been noted are not statistically cignifi- cant The impression is gained from an inspection of table 1 that the large numbers of animals killed ourly in the exporinent aro weighting the final figuroo and detracting from the magnitude of the incidence of tumor It must be noted that this study been an experimental one and that it has been necessary to make observations by killing animals for study at regularintervals It appears that by means of a rather rather involved statisti cal procedure the disproportionate weighting of the values by the data of the early observations can be eliminated This procedure will be carried out after final observations have been made since it is too laborious an undertaking to be performed at the present stage when the values obtainobtaeined d would be only of academic interest Quebc sbestos Mining Assoc 1st Interim Report 5-7-52 o It may he noted in pessing that if the over incidence of tumor anong the exposed animals had been only slightly greater per cont rather than 9.5 per cent difference between the incidence for the , exposed group and for the control group would have been statistically significant An experiment concerning the influence of inhaled asbestos dust upon the incidence of pulmonary tumors in mice has been in progress for 14 months Analysis of the results of this experiment reveals that thus fab after the animals have been exposed to asbestos dust for 14 months the dust has not exerted an influence of a degree sufficient to cause statistically significant alterations in the incidence of pulmonary tumors The of final phases of this exporiment will be of great importance since at the nost rocont period when animals were killed for study the incidence of tumors had increased and the increase seems to be more pronounced among the animals exposed to asbestos dust than among the exposed control animals AJV je 280 1st 1 in Mining 09070 MinMiningin MininggMining Assoc Mining Assoc Assoc Assoc Report 5-7-52 Lf Tablo i Occurrence of Pulmonary Tutors in Mice Exposed to Asbostos Dust and in Mico Not Exposed to Dust ; Strain of nice Strain C Tuner susceptible strain of nice Period of observation mouths NJORA NJORA NJORA Mumber of nico with tumor in the groep exposed to asbestos dust Hala 0 Penalo Number exmined Incidence of tumor Strain C57 Black Tumor susceptiblo - 0 strain of mice 20 14 Number examined Incidence of turer warn. 94 129412.36 129412.36 129412.36 129412.36 129412.36 0 2 OHN 585 : : : 5.95 5.95 5.95 Number of mice with tumor in the control group not exposed to duot llala Perklo 0 1 oO oO oe Oogo 20 69 Incidence of terror in both groups | 27179 27179 27179 -9.58 10181 10181 Zy Mclignant Mclignant Mclignant Mclignant Mclignant tt . wutts ae * a. . 0 0 Table 2 .
