SODATAANALYSIS~~~ 0000
The data from combustion analysis of liver for total organic fluorine are attached. The liver at 48 hours post dose indicates that only the 250 mg/kg dose of T-6053
resulted in significant total organic fluorine in the liver (35 ug in total liver). This is 0.04% solution of an aqueous solution in which the perfluorooctanesulfonate salt is
about 20%. This corresponds to a dose in terms of perfluorooctanesulfonate salt as about 0.02 mg/kg. (The control value is estimated to be about 15 ug total in the
whole liver, but the value is below the quantitation limit and is extrapolated from

The samples are burned in the Dohrman at 950 C using between 8.1 and 8.2 grams of the liver. The gas is collected in 1.0 mL of 1:1 TISAB/Milli-Q waterthen an additional 2 mL of 1:1 TISAB/Milli-Q is added to allow for sufficient volumeforSkalar analysis. The samples are then analyzed on a Skalar Segmented Flow Analyzer using the lon Specific Electrode (ISE) Method.

before the potential between the ion selective electrode and the reference electrode is measured. The signal is amplified and related to the fluoride concentration. The instrument was calibrated in the ranges of 8.815 - 8.15 ppm and 8.15 - 1.58 ppm fluoride. The standard curve for the high range was plotted using the inverse logarithm option. The standard curve for the low range is linear. All standards and samples were then calculated by the Skalar softwareusing these curves. All results below 8.8081 ppm appear on the raw data as #.###4#4,
A quality control standard was analyzed every 18 samples to
check for accuracy and drift. Raw data is taken from the appropriate calibrated range of the Skalar printout and summarized on an Excel spreadsheet. The sfiunbaslerqeuseunlttsdairluetiaodnjsu.sted for the collection volume and any
