Tho test wan conduoted f o r 3n Company for 96 hours from September 18 to 22, 1995, at T.R. nilbury Laboratorian, Inc. in Xarblehead, Haasachusetta. It was conducted by Poter m a l s k i , Joanno IUgaZu, Jacqueline Neviun, Keven Stevenu, Peter NeViUE, Robert Boo& and Timothy Ward trccording to the protocol developed for T.R. wilbury study Number 890-TU. N 2 8 0 3 - 2 uae supplied by the sponaor.
The test was performed under static conditions with five concentration. of test substance and a dilution water control at a temperature of 24 t 2.C. Nominal c!oncentrationr of N2803-2 were 0 (control), 0.99, 2.0, 4.0, 8.0, and 16 mg/L. Tho dilution water WAS aterilo enriched media adjusted to a pH of 7.5. Cell counts were made daily with a hemocytometer.
Exposure of algae to the test substance for 96 hours resulted in a median effective concentration ( E C 5 0 ) of 2.9 mg/L N2803-2, (95% confiderce limits = 1.0 to 7.7 mg/L) when the value war calculated using the number of algal cellr/ml, and 8 . 4 mg/L (95% confidence interval = 5.9 to 14 mg/L) when the value was calculated using the average specific growth rate. The 96 hour no observed effect concentration (NOEC) in 0.99 mg/L and the lowert obaerved effect concentration ( 1 . 0 ~ ~i r) 2.0 mg/L when calculated uning the number of celldm1 and the N O W is 2.0 mg/L And the LoEC is 4.0 mg/L when calculated using tho growth rate.

The a m p l e of N2803-2 (T.R. Wilbury Laboratories Sample Number 520) wan delivered to T.R. wilbury Laboratoriea on August 14, 1995 in a CardSobrd box at smbient temperature. It waa contained in a 50 m l glass jar. The label attached to the jar incXuded the following information:
"N2803-2, see HSDS".
N2803-2 (a clear liquid) was supplied by 3n Company, st Paul,
Minnesota. Prior to use the test substance was atored in the dark at room
temperature. Teat aubstance stock solutions were formulated in dilution
water without the use of a solvent. The test substance was asnumed to
have a purity of 100% active ingredient and to be stable under storage w d

The definitivo toxicity teat Warn porformed from September 18 to 22, 1995, according to T.R. Wilbury Protocol 890-TH (Growth and Roproduction Toxicity Teat with N2803-2 and the Ireahwater Alga, s e l o t u ~ t r u n , caprfcornutum). The protocol, which waa signed by the study director on Auguat 16, 1995, ia baaed on procimduree of the U.S. Environmental Protection Agency (1993). A sununary of the test system is presented in Table 2.
The teot was conducted at a target temperature of 24 f 2-12with five concentrations of test substance and a dilution water control. Nominal concentrations of the test substance meret 0 mg/L (control), 0.99, 2 . 0 , 4.0, 8.0, and 16 mg/L. A 160 mq/L stock solution was formulated on September 18, 1995 by adding 0.080 g N2803-2 to sterile enriched media rnd adjusting the final media volume to 500 ml. Appropriate aaJunts of this stock solution were added directly to dilution water to formulate teat media.
Algae were distributed among three replicates of each treatment at the rat0 of 10,000 cells/ml. The test vas parformed in 250 ml glass Erlenmeyer flasks that contained 50 ml of test solution. Test vessels were randomly arranged on a rotary ahaker adjusted to 100 rpm in an incubator during the test (a random numbers table was used to select the location of each vessel). A 24 hour light and 0 hour dark pbotoperiod was automatically maintained with cool-whit.e fluorescent lights that provided a light intensity of approximately 380 footcandles.
The number of algal cells/ml in each test vessel and the occurrence of relative size differences, unusual cell shapes, colors, flocculations, adherence of cells to test containers, or aggreqation of cella was determined visually by means of direct microscopic examination with a hemocytometer. Cell count. were made and recorded daily during the 96 hour test. ITILTIITICAL NXTBODS: The averago rpocific growth rato wai calculated as tho natural log
of the numbor of cells/ml at 24, 18, 72, and 96 hourrn d n u m tho natural log of the number of cells/ml at 0 hour. divided by the exporure poriod. Tho percent change from tho control waa calculated by mubtracting the treatment average specific growth rato from tho control averago rnpecific growth rate, dividing the differenco by tho avorago rnpecific growth rate in the control, and multiplying that valun by 100.
Results of the toxicity teat were interpreted by standard atatistical techniquer. The probit method wam uaed for calculating 24, 48, 72, and 96 hour EC values (and confidence intorvals). The probit method was use despite the occasional caut.Lonary note because EC50 values computed by thirn method were similar to valuos ccmputod with other methods (stephan, 1983). The calculations were performed using both the number of cella/ml and the averago apecific grovth rates. The na observed effect concentration (NoEC) was determined using a par-tric one-way analysis of variance (ANoVA) and the number of celln/ml and the average specific growth rate in each test vessel at the end of the test.

Biological data generated by the acute toxicity tomt with N2803-2 are premented i n Tables 3 and 4 . The growth curve for algae exposed to
the test aubstance for 96 hours in presented in lfgure 1 (the mlope of the
dome-remponme curve i s 4.1). The 24, 48, 72, and 96 hour tC10, ECSO, and GC9O value6 for algae exposed to N2803-2 &re premented in Table 5.
- - Expomure of algae to the test aubmtance fer 96 houra resulted in a median
effective concentration (EC50) of 2.9 mg/L N2803-2, (9S% confidence limits 1.0 to 7.7 mg/L) when the value van calculated using the number of algal
cells/ml, and 8 . 4 mg/L (95% confidence interval 5.9 to 14 mg/L) when the value warn calculated using the average mpecific growth rate. The 96 hour no obaerved effect concentration (NOEC) is 0.99 mg1L and the lowest observed effect concentration (LOEC) is 2.0 mgfL when calculated using the number of cellm/ml and the NOEC i a 2.0 mg/L and the LOEC is 4.0 m g / ~when calculated using the growth rate.
The determination of whether toxic effects were algistatic or algicidal vas performed at the conclusion of tbe toxicity test. A 0.5 ml aliquot of media from each 16 mg/L veasel vas transferred to a vessel containing fresh media vithout N2803-2 and incubated for 144 hours under test conditions. The number of cellm/ml equalled 1.90,OOO cells/mL, indicating that the effect of N2803-2 at th.is concentration vas algistatic rather than algicidal.
