Il - TEST ANIMALS QFA rats (Sprague-Dewley derived) of SPF quality bred by IFFA CREDO (Lea Oncina G9210 L 'ARBRESLE) are used for this study, 12 - HUSBANDRY All animals are Kept in groups of 5 in cages measuring 43 x 30 x 18 cm on sterile dust free sawdust. Food (SOURIFFARAT) and water aro freely available. These conditions are the same as those in the breeding centre providing the animals; 2 days before the start of the experiment thn animals are placed in their respective groups. Ora 007 TEST ' FOR THE DETERMINATION OF O'CUIAR IRRITATION IN THE RABBIT (following the rcconx&rtdations o f the Journal Official de la Rpublique Franaise o f 21/4/71 and 5/6/73) 1/ OBJECTIVE The test Is used to determine the degree o f irritation resulting from a test substance introduced into the conjunctival sec of the eye of the rabbit. 2/ EXPERIMENTAL PRQT0C01 2.1/ ANIhALS USED Each test requires six traie albino"New Zealand white" rabbits weighing about 2.5 kg at the start of the trial. The animals will have bnen vaccinated against myxomatosis. Care is taken to select only those animals which have e normal eye condition, any with ocular lesions being rejected. 2.2/ HOUSING AND FEEOING The rabbits are kept either in individual cages measuring 500 X 540 X 31S mm, or in restraining devices. Th9 animal house is air-conditioned. 200 g of food is provided per animal per day in the form of granules . (Granuls Lapin "entretien" SANDERS). W.'iter is provided "ad libitum". 2.3/ DOGES ET MODE D 'ADMINISTRATION OU PRODUIT [quantit instille dans le sinus conjonctival de l'oeil - Produit tudi l'EAT PUR, SANS RINAGE ; - En prsence de lsion importante (ex. : opacification cornenne), le produit est de nouveau tudi l'tat pur, mais avec un rinage : 30 secondes aprs 1'irsti1 lation et si ncessaire en dilution dans un solvant appropri (non irritant), sans rinage puis ventuellement avec rinage. 2.4/ MODE OPERATOIRE Chaque lapin est plac dans une bote contention afin de l'irrmobiliser. Le produit tudi est introduit dans le sinus conjonctival de l 'oeil gevicho, l'ooil droit servant de tmoin. Les paupires sont maintenues closes pendant quelques secondes, en effectuant un lger massage, et en vitant toute perte de produit. On laisse les animaux dans leur bote contention pendant 10 heures puis on les remet dans leur cage. N.3 . : En cas de rinagef.vJ, l'excs de liquide est pong avec une gaie strile. 2.3/ LECTURE DE L'IRRITATION OCULAIRE Pour valuer l 'irritation oculaire, les animaux sont placs dans une bote contention. Des lectures sont effectues par rapport l'oeil t/mcin, une heurt aprs l'application du produit, puis J1 (24 heures), J2, J3, J4. J7 ut enfin, en cas d 'irritation persistante, chaque semaine pendant une priode maximum do 5 semaines. L'observation visuelle de l'tat de la corne est pratique l 'aide d 'un ophtalmoscope qui permet galement, par son jeu de lentilles, d'observer distinctement l 'iris, la pupilc* at le cristallin. Un examen oculaire complmentaire peut tre effectu avec une lampe & f o n t e (Waag-Streit). (j V i.' Does notcontain TSCACBi 2.3/ OOSE AND ADMINISTRATION OF TEST SUBSTANCE (amount instilled into the conjunctival sac of the eye - - Substance tested UNDILUTED, WITHOUT RINSING t - When important lesions are found (eg. corneal opicification) the test substance is re-tested in an undiluted state but with rinsing 60 se conds after instillation and, if necessary, diluted with an appropriate non irritant solvent first without rinsing, and subsequently with r i n sing if required. 2.4/ METHOD CP ADMINISTRATION OF TEST PRODUCT Each animal is Immobilised in a restraining box. The test substance is then introduced into the a n i m a l 's left eye, the animal's right eye serves as a control. The eye lids are held closed for several seconds, giving a slight massage and avoiding loss of the substance. The animals are Kept 'restrained for 16 hours and then placed in their cages. N.B. When rinsing($J takes place, excess liquid is wiped away with a sterile gauze. 2.5/ READING THE SITE OF OCULAR IRRITATION To make a reading of the site of irritation, the animals are placed in restraining boxes. Evaluations are made by comparison with the control eye. The readings are made one hour after the administration of the test substance, then at 24 hours after administration, 2 days, 3 days, 4 days and 7 days after administration. When the irritation is persistent, rea dings ore made each week over a maximum period of .5 weeks. Observations of the condition of the cornea are made with the naked eye and also with the aid of an ophthalmoscope. The ophthalmoscope may also be used to observe the iris, pupil end lens. Primary irritants are those substances which, with a single application, cause an inflammatory cutaneous orthoergic reaction. The reaction is localised at the point of contact end appears within 24 hours of application.
