These reports present the results of the acute aquatic toxicity testing conducted by an independent laboratory at the request of the TRP. These studies were conducted according to existing OECD test guidelines and in accordance with the provisions of the appropriate Good Laboratory Practices (GLP) regulations. The physical-chemical characteristics of the test substance indicate that the test substance is a "difficult" substance, as indicated in OECD Monograph 23 (Guidance Document on Aquatic Toxicity Testing of Difficult Substances and Mixtures, OECD 2000), and the experimental designs selected for the acute aquatic toxicity testing were reflective of this observation. Common practices for testing these types of materials, and for evaluating the results from tests of these types of materials, were used during the conduct of the studies and the preparation of the study reports. The TRP and its member companies believe that the data gathered from these studies are consistent with the hypothesis that the 8-2 Telomer B Alcohol is of low to medium concern in terms of its potential for causing acute effects on aquatic species. OECD 202 I Page 7 of 31 Project-No. 020109CK Study-No. DAI84111 1 Summary In the acute immobilisation test with Daphnia magna (STRAUS) the effect of the saturated solution as limit concentration of the test item 1-Decanol, 3,3,4,4,5,5,6,6,7,7,8,8,9,9,10,10,10-heptadecafluoro- (Batch number P00/001), was determined according to OECD Guideline 202 I. The limit test was conducted from 2002-04-03 to 2002-04-05 at D r .U .N o a c k -La b o r a t o r iu m f r a n g e w a n d t e B io lo g ie D-31157 Sarstedt. A coating period of 14 days was carried out. The preparation method was chosen with regard to the test item properties and in accordance with the relevant guidance documents (OECD 2000, BAUA 2001). The study was conducted under static conditions over a duration of 48 hours. 20 test organisms were exposed to the saturation concentration and control. The saturation concentration was analytically verified by GC-analysis (for details see part 8). The mean measured concentration of the saturation concentration was 0.16 mg/L. In the tested saturation concentration of 0.16 mg/L (mean measured concentration) no biologically significant effect was determined. The ECso-value of the reference item at 2.1 mg/L after 24 h was within the prescribed concentration range of 1.0 to 2.5 mg/L according to AQS, DIN Guideline 38412 L 30. Water quality parameters of pH-value and dissolved oxygen concentration measured at 0 and 48 h were determined to be within the acceptable limits. After phase separation 20 mL per test replicate were transferred from the clear liquid phase to each replicate and covered with glass tops. Dilution water was used for the preparation of the coating solution. Coating period The readily filled and covered beakers were stored at nominally 1 8 -2 2 C, 1 C constant for 14 days. At the end of the coating period the beakers were emptied and used for the test. They were obtained by removing the mother animals twice within 22 h. Brood daphnids were maintained in 100 % dilution water at the test temperature for at least 48 hours prior to the test. 20 daphnids divided into 4 (2*) groups with 5 (10*) animals per test vessel were used. Control replicates without test item were tested under the same conditions as the test groups. All test beakers were placed in an incubator (light/dark cycle 16/8 for 48 h at 20 1 C). The immobilisation of the daphnids was observed after 24 and 48 hours of exposure. The reference item was tested in five concentrations within the concentration range 0.58 to 5.8 mg/L. Observations, measurements and evaluation were carried out as described below. Prior to test start the water parameters of the dilution water (pH-value, dissolved oxygen concentration, temperature, conductivity and total hardness) were measured. At the beginning of the test the water parameters (pH-value, dissolved oxygen concentration) were measured in one additional replicate per concentration and control. ECso-value was only calculated for the reference item. ECso-value was calculated via probit analysis. Probits according to W eber (1986). Calculation of the confidence intervals was carried out using standard procedures according to B reitig & TOm p lin g (1982). SigmaPlot rel. 2000 (2000), Spss Corporation VALIDITY CRITERIA The test is unacceptable if more than 10 percent of the control organisms are immobilised during the 48 h test period. The control daphnids should not be trapped on the surface of the water. The EC50 after 24 h of the reference substance should be in the range from 1.0-2.5 mg/L. The dissolved 0 2 concentration at the end should be > 5.21 mg/L (60 % of the air saturation value at the temperature used). OECD 202 I Page 14 of 31 Project-No. 020109CK Study-No. DAI84111 DETERMINATION OF THE TEST ITEM DEVIATIONS FROM THE GUIDELINE DEVIATIONS FROM THE STUDY PLAN REPORTING ARCHIVING The test concentration level and control was analytically verified by gas chromatography with mass spectrometric detection (m/z = 463 and 395) at test start and test end. Additional replicates were set up for the analytical work. None Purity was C8-perfluoroalkylethanol 99.3 % (ala). LOD and LOQ were 0.009 and 0.027 mg/L, respectively. These deviations had no impact on the quality and integrity of the study. The final report is issued as paper copy (6 originals) and an electronic record (1 original). OECD 202 I Page 15 of 31 Project-No. 020109CK Study-No. DAI84111 4 Results 4.1 Solubility of the test item The test item was clearly dissolved throughout exposure. 4.2 Results of the definitive test The percentage immobility was determined in the tested limit concentration and the control after 24 h and 48 h. The immobilisation rates in % after 24 h and 48 h of exposure are given in Table 1. There is no biologically significant effect (10 %) neither in the tested limit concentration nor in control group. 4.3 Results of the reference item The percentage immobility for the reference item was determined after 24 h. The EC10o value was determined directly from the test results. The EC10 and EC with 95 % confidence interval (Cl) were determined by probit analysis according to standard procedures. EC- values after 24 h of the reference item in mg/L were as follows: ECTM : 1.5 ECso : 2.1 (Cl 1 .8 -2 .3 ) EC100 : 5.8 The EC50 - value of reference item potassium dichromate after 24 h is within the prescribed concentration range of 1.0 - 2.5 mg/L of validity criteria according to AQS: DIN Guideline 38412 L 30. 4.4 Water parameters The water parameters (dissolved oxygen concentration, pH-value) of test and control groups at the beginning of the test in one replicate per concentration and control and in all replicates after 48 h are listed in Tables 2 - 3 . pH-value, oxygen concentration, temperature, conductivity and total hardness of the dilution water were measured prior to test start. The validity criteria were fulfilled: In the control group no Daphnia were immobilised or trapped on the surface of the water. Two stock solutions of the standard of 200 mg/L were prepared in terf.-butyl methyl ether, diluted (9 concentrations each) and used for calibration. For calibration range see part 8.2.1. 40 mL of the sample were extracted with 4.0 mL ferf.-butyl methyl ether. After phase separation the extract was injected directly into the GC/MS. METHOD VALIDATION Peak symmetry Repeatability Accuracy Quantification of the test item was calculated by peak area of the masses 69 + 395 + 463 (m/z) based on the external standards. The calibration procedure and the principle for the determination of detection and quantitation limits were adapted from DIN 32645. The method was validated as described below: Peak symmetries were calculated from chromatograms of analytical standards and test item according to the following equation: T = Wo.os / (2-f) T = Peak symmetry W0.05 = Peak width at 5 % of peak height [mm] f = first sector of peak width ( divided by the perpendicular line from the peak maximum ) [mm] 5 sub-samples of the highest and lowest concentration prepared from a single homogenous sample were analysed. Mean values, standard deviations and variations of coefficients were calculated. 5 samples with 40 mL water were spiked with an amount of 8 pg 1Decanol, 3,3,4,4,5,5,6,6,7,7,8,8,9,9,10,10,10-heptadecafluoro- (80 pL of 100 mg/L) to give a final concentration of 200 pg/L. The calibration curve and chromatograms obtained for standard and test item are given in Fig. 1 - Fig. 6. For limits of determination and quantification see part 8.3. 8.2.2 Quantification Limit and Detection Limit The lim it of quantification (LOQ) and limit of detection (LOD) were calculated using the blind value method described in DIN 32645 "Nachweis- und Bestimmungsgrenze". Therefore the second curve (2002-04-10) which covers a much wider range ( 0 . 1 - 1 0 mg/L) was used for the calculations in this test. The concentration calculations using the two different curves do not differ more than 10 % from each other.
