Although experimental studies have demonstrated the carcinogenicity and mutagenicity of VC/PVC in general, the epidemiologic studies available for review do not include an assessment of carcinogenic risk among humans exposed to these chemicals. This conclusion is based on the observation that the majority of cohort studies reviewed lacked sufficient statistical power because of small sample sizes. Further, in epidemiological studies, individuals were not followed over an adequate period of time during which cancer could become clinically manifest. In this study 6- and 8-vinyl-and -ethynyluridine and -adenosine were prepared by a general procedure involving the palladium-catalyzed cross-coupling of trimethylsilyiacetylene or vinyitributyltin. The introduction of a vinyl group at C-6 of uridine or an ethynyl group at C-8 of adenosine resulted in nucleoside derivatives showing cytostatic activity against several murine and/or human tumor cell lines. Interestingly, 8-vinyladenosine had pronounced selective inhibitory effects on human (Molt/4F and MT-4) versus murine (L1210 and FM3A) tumor cell lines. Six developments occurring in 1974 related to pesticides are reviewed: the "'vinyl*** '"chloride*** case; the regulation of 14 carcinogens under OSHA; the banning of aldrin and dieldrin; cancer among U.S. arsenic "'workers*** ; the relation of neurotoxicity and carcinogenicity; `"human*" poisoning due to chlorinated-p-dioxins; and the encapsulation of pesticides. The findings of cases of hemangiosarcoma of the liver among '"workers'" in polyvinyl chloride demonstrated that there is a need to retest older pesticides and associated materials that have never been adequately assessed on a long-term basis to disclose carcinogenicity. Of the 14 carcinogenic chemicals brought under control on Feb. 11,1974 by the OSHA, only two, beta-propiolactone and ethyleneimine, could be considered pesticidal. They constitute a potential occupational hazard for an extremely small percentage of "'workers'** exposed to pesticides. In 1974 dieldrin and aldrin were established as carcinogens. Large numbers of '"workers"* have been exposed to occupational contact with these carcinogens before. Hemangioendothelioma of the liver was diagnosed in vineyard '"workers'" . Last year there '"leukemia*" was reported in a U.S. '"worker"* using arsenic pesticides. Studies of '"workers"* manufacturing arsenical sheep dip have strongly suggested that lung cancer occurs in higher than expected rates in these arsenic exposure occupations. Pesticides as a class of * compounds appears to strengthen the idea of a relation between neurotoxicity and carcinogenicity. For a time, toxicologic test procedures were not adequately modified to avoid marketing products which contained high toxicity contaminants. A technique has been developed for the microencapsulating of pesticides to provide slow controlled release. Two years of field trials are cited during which the encapsulated methyl parathion diffusing through the pores of the polyamide microspheres maintained levels on crops for much longer than did conventional methyl parathion treatment. Residues of the encapsulating polyamide are inert and nontoxic in certain food crops. Microcapsules can be readily sprayed on as a water slurry. Caution is advised as the "'vinyl*** '"chloride"* case has alerted the health-related professions to the occurrence of unreated monomers and additives in seemingly nonreactive plastics. Even smaller nylon type polymer spheres are available. These could be inspired into the periphery of the lung where release of unreacted monomers and additives would presumably ensue. Ethylene oxide has been produced by a Swedish company since the beginning of the 1940s. This paper describes a cohort study of the mortality and the cancer incidence among full-time exposed workers in ethylene oxide production, a group of maintenance workers with intermittent exposure and a group of unexposed controls. Investigation of the production processes in the building at different times has shown that workers were exposed to ethylene dichloride, ethylene chlorohydrin, ethylene, and small amounts of bis-(2-chloroethyl) ether as well as to ethylene oxide and traces of other chemicals. The full-time exposed cohort shows a considerable excess mortality deriving mainly from increased mortality from tumours and also from diseases of the circulatory system. The cancer incidence study, including living persons with malignancies, showed a significant excess in the full-time cohort. Of the 16 patients with tumours in the two more exposed cohorts there were three cases of leukaemia, six of tumours in the alimentary tract and four of urogenital malignancy. The excess mortality and cancer incidence cannot be attributed to any particular chemical in the production process, but ethylene oxide and ethylene dichloride are the prime suspects. Treatment of 2,4-bis(trimethylsilyl)-5-fluorouracil with either 2-methoxy-3-acetoxytetrahydrofuran or 2-ethoxy-4acetoxytetrahydrofuran in ***1"* , ***2*** -dichloroethane at room temperature using SnCI4 as catalyst afforded cis- and trans-3'-OAc-FT or 4'-OAc-FT, respectively. However, trans-3'-OAc-FT and cis-4'-OAc-FT were the major condensation products. In each case, separation of these cis and trans isomers was achieved by silica gel column chromatography. Treatment of 3'- or 4'-OAc-FT with NH3/CH30H at 5 degrees C overnight yielded the described hydroxylated FT. The cohrt was set up by 226 workmen on 1960 and observed until 1979. No cause of death were statistically significant with p<0.05. However, death from all cause, all cancer, gastric cancer, liver and bilialy tract cancer, lung cancer, "`leukemia*** , hypertension, other heart disease, liver cirrhosis, and accident were higher than expected. Death from all cause, all cancer, liver and bilialy tract cancer and lung cancer were not significantly higher with p<0.05 level, however, these causes were significantly higher than Japanese male with p<0.10 level. Moreover, the relative death rates of both liver and bilialy tract cancer and lung cancer were increased in accordance with the length of VCM exposure. CODEN: BJIMAG ISSN: 0007-1072 LA English AB The results of a cancer mortality study among workers employed in the production of vinyichloride and polyvinylchloride between 1939 and 1977 suggest a significant increase in deaths from malignancies of the lymphatic and haemopoietic tissues. Mortality for tumours of the digestive organs, respiratory system, bone and connective tissues, brain, and skin are also greater than in the general population. There were no registered cases of liver angiosarcoma in the study cohort during the follow up period. The risk of cancer was highest among the workers exposed to concentrations of VC of 300 mg/m3 and more who had worked at the plant for 15 to 19 years. The relatively high number of leukaemias and lymphomas in the study group and the absence of liver angiosarcomas probably reflects specific carcinogenic action of different doses of vinyichloride. We have recently developed two ultrasensitive methods for determining the molecular dose of these adducts in cellular DNA. In both methods, purified DNA was first enzymatically digested to 2c-deoxyribonucleotide 3c-monophosphates. Etheno-modified nucleotides were then separated from normal nucleotides in one of two ways: either by reverse phase, ion-pair HPLC coupled with 260 nm UV detection, or by immunoaffinity chromatography using reusable microcolumns containing specific monoclonal antibodies coupled to Protein A-Sepharose. Although there is concern that our environment is deteriorating, there is little evidence that this is leading to an increased cancer burden as, although rates in the elderly have been increasing, rates in younger people are declining. It is estimated that only about 5% of cancers are due to environmental factors and more than half of these are due to the geophysical properties of the environment, rather than manmade pollutants. Hazards to which people are exposed at work do contribute to increased cancer rates. Once such a hazard is identified then legislation is used to limit, or eliminate, exposure to it, both at work and in the environment. This paper describes certain specific hazards, but concludes that the largest contribution to cancer prevention would be a reduction in smoking rather than any environmental measure. In accordance with the Michigan Environmental Response Act, the state provided the affected residents with bottled water for drinking purposes. Risk assessments for carcinogens (benzene, 1.2-dichloroethane, dichloromethane, trichloroethylene, and tetrachloroethylene) were formulated. The total excess cancer cases attributable to the consumption of contaminated water from these wells were estimated to be 14 per one million persons. The maximum concentration of chloroform in the city public water supply was 26 ug/L. The high concentration of chloroform was the result of chlorination of the unfiltered river water by the public water facility. The relative health risk from the consumption of chlorinated surface water from the public water supply system would be approxfmatety 4.3 times greater compared to that of .consuming groundwater from the contaminated private wells. The affected residents were given three options: (a) continue consumption of bottled water; (b) connection to the existing public water supply system; or (c) construction of deep water wells. The citizens voted , for the second option of connecting to the public water supply system and voluntarily accepted the relatively higher health risk. The State of Michigan later proposed to further improve the water purity by upgrading the public water supply system by the incorporation of a filtration plant. Unexposed chem. industry workers had slightly high rates. The standardized mortality rates (SMR) for malignancies of lymphatic and hematopoietic tissues were significantly high in exposed workers. PVC processing workers had esp. elevated SMR for brain malignancies. All workers had elevated SMR for ischemic heart diseases and acute cardiac infarctions.
