INTRODUCTION ToxicittyestswithMicroto& providea rapidscreeningmethod and can be reliablperedictorosftheinhibitory effectsofcontaminantson microbiaslystems.
Thistoxicitsytudy measured the lighotutputofbioluminescentbacteriaovera relativeslhyortperiod(5,15,and 30-minutes)inthe presence ofdifferenctoncentrationosf P3025.

Test solutionwsere preparedas Water-Accommodated Fractions(WAFs). Four stocksolutionosf 250,500, 1000,and 2000 mg/L were preparedinMilliporMeilli0 water. Each was capped and allowedtomix overnighti,nthedark,atambient room temperature.Test substancewas apparentlynotcompletelymisciblewith water. A few whiteparticulatewsere observed ineach stocksolutioanftermixing.
Justpriortotestinitiatiaon1,0 mL aliquotoftheclearliquipdortionofeach stocksolutiownas removed and each was osmoticallaydjustedwith200 mg reagentgrade NaCi. Each initicaolncentratiownas dilutedby 50% duringthe course ofthetest.

Measurements Procedure
Lightreadingswere taken upon reconstitutionfthe Microtox@ bioluminescentbacteria, and then 5,15,and 30-minutesaftertestsubstance was added tothe bacteria.All readingswere captured electronicalvliyacomputer interfacweiththeMicrotoxS instrument.
The testwas conducted ingeneralaccordance withtheAzur EnvironmentalMicrotox@ Basic Test.
CALCULATIONS
The reportedvalues are based on nominal concentrationsinthe testmedia atthebeginningofthetest,The concentrationstestedand the correspondingresponse data derivedfrom the toxicittyestare used tocalculate,it possible,the medial effectiveconcentrationE,C50 and the 95% confidenceintervably standard logistic regressionmethods provided inthe Azur Environmentalsoftware.The EC50 isdefinedas the concentration estimatedtoinhibitthe rateof bioluminescenceby 50% as compared tothe controlsa,fter5,15, and 30-minutes of exposure tothe testsubstance.

RESULTS The percentreductioninlightoutputand the median effectivceoncentrationE,C50, obtainedafter5,15 and 30minutes exposure tothe testsubstance P3025 are summarized inTable 1. The median effectivceoncentration3,0-minute EC50 and 95% confidenceintervawlas reportedas 350 (240-490) mg/L.
ACCEPTABILITY QRITERIA Thistoxicittyestmeets qualitycriteriparovidedby Azur EnvironmentalCorporation:
1) The 95% confidencefactorforthe EC50 was <1.3 forthe 5 and 15-min exposure periods. Itwas > 1.3at30-min,,however,thiswas probablydue tocomplete inhibitioantthe highest concentratiotnested(1000 mg/L) .
2) The coefficienotfdeterminationwas > 0.9 forthe 5 and 15-min exposure periods. Itwas < 0.9 at30-min;however, thiswas probablydue tocomplete inhibitioantthe highest concentrationtested(1000 mg/L)
3) The correctiofnactorwas > 0.7at5-min and > 0.6at15 and 30-min. This new versionofAzur EnvironmentalCorporationsoftwaredoes notprovidethe correlatiocnoefficient.
