VIII. HETBODS AND MATERIALS

The sample of N2803-4 (T.R. wilbury Laboratories Sample Number 522) was delivered to T.R. Wilbury Laboratories on August 14, 1995 in a cardboard box at ambient temperature. It was contained in u1 50 ml glass jar. The label attached to the bottle included the fo1:lowing information: "N2803-4, (see U S D S ) " .
N2803-4 (white powder) was supp1.ied by 3M Company, St Paul, Hinnesota. Prior to use the test substance was mtored in the dark at room temperature. The test substance was asnumed to havei a purity of 1001 active ingredient and to be stable under storage and testing conditions. Unumed teat substance will be returned to the sponmor.

Water used for acclimation of test organisms and for all toxicity testing was sterile enriched media (U.S. EPA, 1978; T.R. Wilbury standard Operating Procedure number 6 ) adjusted to a target pH of 7.5 with 0.1 N HCl prior to use. characterization of a representative sample of water used to formulate media is presented in Tabls 1.

Algae used for the test (Selenastrum caprfcornutum, VTEX 1648) vas from a c u l t u r e originally procured f r o m t h e culture COllleCtiOn of Algae at the University of Texas at Austin and delivered to T.R. wilbury Laboratorios on April 11, 1995. The definitive toxicity test was performed from September 22 to 26, 1995, according to T.R. nilbury Protocol 896-TH (Growth and Reproduction Toxicity Teat with N2803-4 and the! Freshwater Alga, Selonastrum caprfcornutum). The protocol, which waii signed by the atudy director on August 16, 1995, i s based on procedures of the U.S. Environmental Protection Agency (1993). A summary of the test system ia pre8ented in Table 2.
The test was conducted at a target temperature osf 24 f 2-12 with five concentrations of teat substance and a dilution water concrol. Nominal
concentrations of the test substance were: 0 mg/L (control), 62, 130, 250, 500, and 1,000 mg/L. A 1,000 mg/L stock solution was formulated on September 22, 1995 by adding 0.5 g !r280:1-4 to sterilfl enriched media and adjusting the final media volume to 500 ml. Appropriate amounts of this atock solution were added directly to dilution water to formulate teat media.
Algae were distributed among threa replicates cbf each treatment at
the rate of approximately 10,000 cellslml. The tefrt was performed in
250 ml glass Erlenmoyer flasks that contained 50 ml. of test Solution.

Test vessels were randomly arranged on a rotary shaker adjusted to 100 rpm
in an incubator during the test (a random numbers table was used t o select
the location of each vessel). A 24 hour light and 0 hour dark photoperiod
waa automatically maintained with coo:l-white fluorescent lights that
provided a light intensity of approximately 400 footcandles.
The number of algal cells/ml in each test vessel. and the occurrence of relative size differences, unusual cell shapes, cdars, 'locculations, adherence of cells to test containers. or aggregation of cells was determined visually by means of direct microscopic examination with a hemocytometer. cell counts were made and recorded daily during the
96 hour test. The determination of whether toxic effects were algistatic or algicidal was performed at tho conclunion of the t o x i c i t y test (0.5 m l

The avmrage a p e c i f i c growth rate was c a l c u l a t e d as thm n a t u r a l l o g
of t h e number of cells/ml A t 24, 48, 72, and 96 hours minus t h e n a t u r a l l o g o f t h e number of cells/ml a t 0 hours d i v i d e d by t h e e x p o s u r e p e r i o d . The p e r c e n t change from t h e c o n t r o l was c a l c u l a t e d by s u b t r a c t i n g t h e treatment average s p e c i f i c growth rate from t h e c o n t r o l average s p e c i f i c growth rate, dividing t h e difference by t h e average s p e c i f i c growth rate i n t h e c o n t r o l , and m u l t i p l y i n g t h a t v a l u e by 100.
R e s u l t s of t h e t o x i c i t y t e . i t were i n t e r p r e t e d by s t a n d a r d s t a t i s t i c a l techniques. The binomial/interpolat.ion or p r o b i t methods ( s t e p h a n , 1983) were used f o r c a l c u l a t i n g EC10, E:C50, and EC90 values. A l l c a l c u l a t i o n . were performed u s i n g t h e number of cells/ml and t h e averagm s p e c i f i c growth r a t e s and nom.ina1 c o n c e n t r a t i o n s o f N2803-4. The no observed e f f e c t C o n c e n t r a t i o n (NOEi:) was d e t e r a i n e d rrsing a p a r a m e t r i c one-way a n a l y s i s o f v a r i a n c e (ANOVA) or a non-parametric Kruskal and Wallis' test and t h e number of c e l l s h l and t h e average s p e c i f i c growth rate in each test vessel a t t h e end of the test.

no insoluble material waa notod in any test vessel during tho test. The algal population in the control vosae:!s grew wall, resulting i n an average of 1,407,000 cells/ml after 98 hours (Tablo 3). Water quality throughout the test waa within acceptable limits (Append.Lx A). Tho range of incubator temperatures was 23.5 to 23.7.C (T.ble A . 1 ) . The pE of test media (Tablo A.2) was slightly affected by the teat substance only at 1,000 mg/L at the beginning of the tost.
Biological data generated by the acute toxicity t'est with 12803-4 are presented in Tables 3 and 4. The growth curve for algae exposed to the test substance for 96 hours is prosanted in Figure 1 (;thealope of the dose-response curve is 2.5 based on cells/mL at 96 hours). The 7 2 and 96 hour EC50 value!, for algae exposed to N280.1-4 are presented in Table 5. The 96 hour EC50 (and associated 95 percent confidence 1fDiits) 1s 310 mg/L ( 2 8 0 and 350 mg/L) when calculated using the number of cells per ml, and greater than 1,000 mg/L when calculated using the average specific growth rate. No effects (size differences, uiiusual cell shapes, colors, flocculations, adherence of cells to test container!,, 01: aggregation of cells) were noted. The 96 hour no observed affect concentration (NOEC) is 62 mg/L and the lowest observed effect conc:entration (LOEC) is 130 mg/L 32803-4, when calculated using the number cells/ml, and the NOEC is 500 mg/L and the LOEC is 1,000 mg/L. when calculated uaing the average specific growth rate.
The determination of whether toxic effects were1 algistatic or
algicidal was performed at the conclusion of the toxicity test. A 0.5 m l
aliquot of media from each 1,000 mg/L vesso:L was transferred to a vessel
containing fresh media without N2803-4 and incubated for 72 hours under
test conditions.
Algae increased from <10,000 cella/ml to
642,000 cells/mG, indicating that the effect of N2803-4 at this
concentration was algistatic rather than algicidal.

111. PROTOCOL hMENDMUTS AND DWIATIOUS
The protocol was amended to specify the nominal concentrations for the definitive study. No other amendments were made to the protocol. PROZDCOL DEVIATIONS: Two nominal concentrations were slightly diliferent than the ones listed in the protocol. This deviation did not affect the outcome of this test and no other deviations were mado from the protocol.
