To better understand the cellular mechanisms of liver
carcinogenesis, cells isolated from a liver ***tumor*** , in a
worker occupationally exposed to vinyl chloride for 23 yr, were
established in culture. These cells, BM 2.2.1, which could be
expanded during 30 passages, were characterized. They exhibited a
morphol. similar to that of transformed fibroblasts. They were
neg. for desmin, smooth muscle myosin, cytokeratin 19 and von
willebrand factor, pos. for vimentin and showed stress fibers
strongly labeled for .alpha, smooth muscle actin (.alpha.SMA) with
an important deposition of fibronectin around the cells.
Ultrastructure showed typical myofibroblast-like features (e.g.
microfilaments bundles). Their chromosome no. ranged from 38 to
168 with two populations-, a near diploid one with 41 to 47
chromosomes and a hypotetraploid one with 74 to 87 chromosomes.
Structural aberrations were found, esp. loss of chromosome Y and
double-minutes were obsd. in every mitose. No ***mutations***
were found in codons 12 and 13 of the Ki-ras gene whereas a point
***mutation*** at codon 179 (CAT.fwdarw.CTT) was detected in the
p53 gene. Finally, these cells presented: (1) typical
myofibroblastic features (particularly the expression of
.alpha.SMA), (2) a karyotype suggestive of ***tumoral*** cells,
(3) a mutated p53 gene, suggesting that among the liver mesenchymal
cells, both endothelial and fibroblastic cells (particularly
perisinusoidal stellate cells) may acquire a ***tumoral***
phenotype and participate in the development of so called
angiosarcomas.

AB The case of liver ***sarcoma*** in a 47 yr-old male worker
exposed te vinyl chloride during 22 yr is presented. During that period the patient inhaled about 0.8105 kg of vinyl chloride. It is estd. that the asymptomatic phase of liver angiosarcoma lasted 21 yr. The patient died 4 mo after the occurrence of clin. symptoms.
